Effects of bimolane and probimane on the incorporallon of [3H]TdR, [3H]UR and [3H]Leu into Ehrlich ascites carcinoma cells in vitro

Abstract

By means of liquid scintillation spectrometry. the effects of bimolane (AT-1727) and probimane (AT-2153) on the incorporation of [3H]TdR, [3H]UR and [3H]Leu into the Ehrlich ascites carcinoma cells were studied. The incorporation of [3H]TdR. [3H]UR and [3H]Leu into ECA cells was quickly inhibited after being exposed to AT-1727 and AT-2153, and the rate of inhibition showed a close relationship to the dosage of the drugs. The inhibition recovered after the rcmoval of AT-1727, but the recovering was slow after the removal of AT-2153. This experiment showed that AT-1727 exerted an inhibitory action on DNA synthesis in tumor cells through interfering the metabolism of DNA while AT-2153 through the damage of DNA template of tumor cells.
