Comparison of arrhythmic effects of aconitine and its 5 analogues

Abstract

The acute ip LD50 in mice of mesaconitine (MA), aconitine (AC), beiwutine (BA), hypaconitine (HA), 3-acetylaconitine (AA), and deoxyaconitine (DA) were 0.22, 0.27, 0.42, 0.50, 1.01 and 1.90 mg/kg, respectively.
These 6 drugs iv induced arrhythmias in anesthetized rats. The potencies of these drugs (1/14 of LD50) were BA>MA>AC=AA>HA>DA. BA and DA yielded a modest, while other drugs produced a severe, respiratory depression.
BA 10 microg/kg iv caused arrhythmia in anesthetized rats, yet the drop of blood pressure was not marked. BA in aqueous solution was more steady than AC. It is suggested that BA is superior to AC to produce arrhythmia models for screening of drugs.
