Linear transformation of Michaelis-Menton equation by Lineweaver-Burk's method
Abstract
Mathematical model analysis has demonstrated that the original Lineweaver-Burk equation is very sensitive to low response errors, hence its exacting characteristic may be explained. A modified version of the L-B formula, the dose response ratio method, having the advantage of obviating the double reciprocal, is devoid of the error amplification due to the least square procedure. Results of comparative computation of some experimental data by these 2 euations showed that the error due to the dose response ratio formula was smaller than that obtained in using the usual L-B method. The dose response ratio method is therefore recommended in calculating the receptor dynamics parameters(EA max and KA).
